McCaffrey suffered the injury on a 7-yard touchdown run in the fourth quarter. He had his ankle taped on the sideline but was ruled out and did not return to the game. He underwent an MRI on Monday.
He also did something to his leg in the first quarter and came out of the game, coach Matt Rhule said in the post-game video call with reporters.
He finished the game with 59 yards on 18 carries and two touchdowns. McCaffrey, 24, signed a record four-year, $64 million extension with the Panthers last spring.
There were a number of star players who suffered injuries in Week 2 of the NFL season, including fellow backs Saquon Barkley of the New York Giants, Raheem Mostert of the San Francisco 49ers and David Montgomery of the Chicago Bears.
